EMBO Practical Course Strategies in Macromolecular Structure Determination at 3rd Generation Synchrotron Sources

Introduction
Synchrotron radiation has become a important tool in the determination of the 3-dimensional structure of biological macromolecules. The objectives of this practical course are to give scientists a sturdy foundation in the different methods in macromolecular crystallography specifically employed at synchrotron radiation facilities - especially data collection techniques and phasing methods with anomalous scattering (MAD & SAD methods), atomic resolution studies and automation.
The practical course will consist of:
- Lectures of the theoretical principles and their applications,
- Hands-on tutorials using programs for data integration, substructure determination and phase calculations,
- Practicals on MAD & fixed wavelength beamlines at the ESRF.
The lecture topics will cover:
- General theory and application of anomalous scattering to macromolecular crystallography
- Data collection, integration & processing
- Substructure determination via Patterson and direct methods
- Calculation of experimental phases
- Model building, High resolution studies & Automation
- Novel Methods
