Scope of the course
Multiple- & Single-wavelength Anomalous Diffraction (MAD & SAD) experiments have become the methods of choice for determining the 3-dimensional structures of macromolecules. With the enormous advances in methodology, software and instrumentation, even the weak anomalous scattering signals from sulphur can be exploitated. The objectives of this practical course are to teach these anomalous scattering methods to young scientists who intend to apply MAD, SAD & sulphur phasing techniques. There will be a strong emphasis on handling instrumentation on the beamlines, improving experimental skills in X-ray data collection, and solving crystal structures. Students will have the opportunity to meet the authors of structure determination programs and to collect MAD/SAD data.
